Revisiting the Source of Wilt Symptoms: X-Ray Microcomputed Tomography Provides Direct Evidence ThatRalstoniaBiomass Clogs Xylem Vessels

چکیده

Ralstonia causes wilt diseases by colonizing xylem vessels and disrupting water transport. The dogma is that bacterial biomass clogs reduces the flow of sap due to abundance. However, physiological mechanism disruption during untested. Using a tomato pseudosolanacearum GMI1000 model, we visualized quantified spatiotemporal dynamics wilt. First, measured stomatal conductance leaflets on mock-inoculated wilt-symptomatic plants. Wilted had reduced conductance, as did turgid same petiole wilted leaflets. Next, used X-ray microcomputed tomography (X-ray microCT) light microscopy differentiate between mechanisms disruption: blockage biomass, vascular tyloses, or displacement gas embolisms. We imaged intact plant stems quantify embolized vessels. Embolized were rare, but infected plants with low populations nonsignificant trend more vessel To test whether are clogged wilt, excised after brief dehydration. Most in mock-infected emptied their contents excision, nonconductive abundant 2 days post infection. At onset, when exceeded 5 × 10 8 CFU/g stem tissue, approximately half electron-dense biomass. found no evidence tyloses microCT reconstructions from preserved stems. Extracting multiple interacting root systems using X‐ray microcomputed tomography

Root system interactions and competition for resources are active areas of research that contribute to our understanding of how roots perceive and react to environmental conditions. Recent research has shown this complex suite of processes can now be observed in a natural environment (i.e. soil) through the use of X-ray microcomputed tomography (μCT), which allows non-destructive analysis of pl...

Line-Source Based X-Ray Tomography

Current computed tomography (CT) scanners, including micro-CT scanners, utilize a point x-ray source. As we target higher and higher spatial resolutions, the reduced x-ray focal spot size limits the temporal and contrast resolutions achievable. To overcome this limitation, in this paper we propose to use a line-shaped x-ray source so that many more photons can be generated, given a data acquisi...

Theory of X-ray microcomputed tomography in dental research: application for the caries research

Dental caries, a chronic infectious disease, affects more than 90 percent of all adults in the United States and over 80 percent of all adults in Korea. Historically, caries have been prevalent since the time of pre-Neolithic humans (10,000 BC) with a reported prevalence between 1.4% and 12.1% carious teeth, but it was not until the fourteenth and fifteenth century when a sharp increase in cari...

Differences in Trabecular Bone Mechanical Strength Measured by the X-ray Microcomputed Tomography and Compressive Test

The article describes the concept of measurement of the mechanical strength of trabecular bone dissected from human femoral head. When the studies are performed based on the human trabecular bone, there is a risk that correlation can be not achieved due to high discrepancy in the distribution of the bone density and mechanical properties. The compressive tests and X-ray microcomputed tomography...
